Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.3160G>T (p.Gly1054Trp), citing Ambry Variant Classification Scheme 2023: The p.G1054W variant (also known as c.3160G>T), located in coding exon 18 of the PTCH1 gene, results from a G to T substitution at nucleotide position 3160. The glycine at codon 1054 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
