Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.4007G>T (p.Arg1336Met), citing Ambry Variant Classification Scheme 2023: The p.R1336M variant (also known as c.4007G>T), located in coding exon 23 of the PTCH1 gene, results from a G to T substitution at nucleotide position 4007. The arginine at codon 1336 is replaced by methionine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
