Uncertain significance — the classification assigned by GeneDx to NM_001005242.3(PKP2):c.250C>T (p.Pro84Ser), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the PKP2 gene. The P84S variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The P84S variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The P84S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties, and this substitution occurs at a position that is conserved across species. Additionally, the majority of in silico algorithims (2 out of 3) predict this variant is probably damaging to the protein structure/function.
