NM_018699.4(PRDM5):c.1544G>A (p.Arg515His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R515H variant (also known as c.1544G>A), located in coding exon 14 of the PRDM5 gene, results from a G to A substitution at nucleotide position 1544. The arginine at codon 515 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
