Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.279A>C (p.Arg93Ser), citing Ambry Variant Classification Scheme 2023: The p.R93S variant (also known as c.279A>C), located in coding exon 3 of the POLE gene, results from an A to C substitution at nucleotide position 279. The arginine at codon 93 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
