Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.1741G>C (p.Ala581Pro), citing Ambry Variant Classification Scheme 2023: The p.A581P variant (also known as c.1741G>C), located in coding exon 16 of the POLE gene, results from a G to C substitution at nucleotide position 1741. The alanine at codon 581 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
