This variant lies in the POLE gene (transcript NM_006231.4) at coding-DNA position 4191 through coding-DNA position 4192, deleting 2 bases. Submitter rationale: The c.4191_4192delCT variant, located in coding exon 33 of the POLE gene, results from a deletion of two nucleotides at nucleotide positions 4191 to 4192, causing a translational frameshift with a predicted alternate stop codon (p.Y1398*). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
