Submitter rationale: The c.4693dupT variant, located in coding exon 36 of the POLE gene, results from a duplication of T at nucleotide position 4693, causing a translational frameshift with a predicted alternate stop codon (p.C1565Lfs*35). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
