Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002691.4(POLD1):c.2766_2772dup (p.Val925fs), citing Ambry Variant Classification Scheme 2023: The c.2766_2772dupCCCCTAC variant, located in coding exon 21 of the POLD1 gene, results from a duplication of CCCCTAC at nucleotide position 2766, causing a translational frameshift with a predicted alternate stop codon (p.V925Pfs*31). This alteration is expected to result in protein truncation or nonsense-mediated mRNA decay. However, loss of function of POLD1 has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
