This variant lies in the SI gene (transcript NM_001041.4) at coding-DNA position 1126, where C is replaced by T; at the protein level this means replaces arginine at residue 376 with tryptophan — a missense variant. Submitter rationale: The p.Arg376Trp variant in the SI gene has not been previously reported in association with disease. This variant has been identified in 3/251,002 chromosomes by the Genome Aggregation Database (http://gnomad.broadinstitute.org/). Although this variant has been seen in the general population, its frequency is low enough to be consistent with a recessive carrier frequency. In silico tools predict that the p.Arg376Trp variant is deleterious; however, these predictions have not been tested directly. Using ACMG guidelines, this variant was classified as a variant of uncertain significance (ACMG evidence codes used: PM2_supporting, PP3).
