Submitter rationale: The c.837+45A>G variant in the LDHB gene is in trans with the p.Ser256Asn variant identified in this individual, but has not been previously reported in association with disease and was absent from large population databases, including the Genome Aggregation Database (http://gnomad.broadinstitute.org/). This variant occurs in the 3’ splice region and computational tools do not predict an impact to splicing. These predictions have not been tested directly. Using ACMG guidelines, this variant was classified as a variant of uncertain significance (ACMG evidence codes used: PS3_supporting, PM2_supporting, BP4).
