Pathogenic for Unstable hemoglobin disease — the classification assigned by Centre for Research and Development of Medical Diagnostic Laboratories, Khon Kaen University to NM_000518.5(HBB):c.[70G>T;79G>A]: The mutation was detected in an adult Thai female during prenatal diagnosis of thalassemia and hemoglobinopathies. DNA sequencing analysis of the whole β-globin gene revealed double mutations of Hb E (ClinVar ID: 15161) and Hb Palmerston North (ClinVar ID: 15306) in the same β-globin gene, namely Hb E-Palmerston North, which could be confirmed by multiplex allele-specific amplification. Hb analysis showed an unusually low level of Hb E in heterozygotic form. System stability prediction of the Hb E-Palmerston North expression indicated a weakening αβ contact, resulting in instability and a dramatic reduction of Hb variant (3.4%).
