Likely pathogenic for Colobomatous optic disc-macular atrophy-chorioretinopathy syndrome — the classification assigned by Kasturba Medical College, Manipal, Kasturba Medical College, Manipal, Manipal Academy of Higher Education, Manipal, India to NM_007374.3(SIX6):c.433G>T (p.Glu145Ter), citing ACMG Guidelines, 2015. This variant lies in the SIX6 gene (transcript NM_007374.3) at coding-DNA position 433, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 145 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: A novel stop-gain variant, c.433G>T in exon 1 of SIX6 was observed in homozygous state in the proband. On segregation analysis the variant was observed in heterozygous state in the father and mother. This variant is absent in the gnomAD (v4.1.0) population database and in our in-house data of 3550 exomes. This variant introduces a premature termination codon which may either cause the transcript to undergo nonsense-mediated mRNA decay or lead to the formation of a truncated protein product.
