Uncertain significance for Dilated cardiomyopathy 1JJ — the classification assigned by Petrovsky National Research Centre of Surgery, The Federal Agency for Scientific Organizations to NM_001105206.3(LAMA4):c.2456G>A (p.Arg819Gln), citing ACMG Guidelines, 2015: Heterozygous variant NM_001105206.3:c.2456G>A (p.Arg819Gln) in the LAMA4 gene was found on WES data in female proband (51 y.o., Caucasian) diagnosed with dilated cardiomyopathy. The variant is in The Genome Aggregation Database (gnomAD) v4.1.0 with total MAF 0.000001239 (Date of access 14-11-2024). Heterozygous mutations in LAMA4 associate with dilated cardiomyopathy type 1JJ (MIM number 615235). However, in the absence of the functional studies, we could only classify this genetic variant as a Variant of Uncertain Significance.
