Uncertain significance for Familial Mediterranean fever — the classification assigned by 3billion to NM_000243.3(MEFV):c.1966G>A (p.Glu656Lys), citing ACMG Guidelines, 2015. This variant lies in the MEFV gene (transcript NM_000243.3) at coding-DNA position 1966, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 656 with lysine — a missense variant. Submitter rationale: The variant is not observed in the gnomAD v2.1.1 dataset. Predicted Consequence/Location: Missense variant In silico tool predictions suggest damaging effect of the variant on gene or gene product [REVEL: 0.72 (>=0.6, sensitivity 0.68 and specificity 0.92); 3Cnet: 0.93 (>=0.6, sensitivity 0.72 and precision 0.9)]. A different missense change at the same codon (p.Glu656Ala) has been reported to be associated with MEFV- related disorder (PMID: 11464238). However the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
