NM_032043.3(BRIP1):c.1474-1G>T was classified as Likely pathogenic for Hereditary cancer-predisposing syndrome by Molecular Diagnostics Laboratory, Catalan Institute of Oncology, citing ACMG Guidelines, 2015. This variant lies in the BRIP1 gene (transcript NM_032043.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1474, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: PVS1, PM2_Supporting c.1474-1G>T, located in a canonic splicing site of the BRIP1, is predicted to alter splicing, probably causing the skipping of exon 11 (r.1474_1628del). This alteration is expected to result in loss of function by premature protein truncation and nonsense-mediated mRNA decay (PVS1). It is not present in the population database gnomAD v2.1.1, non cancer dataset (PM2_supporting). To our knowledge, neither relevant clinical data nor well-stablished functional studies have been reported for this variant. Also, the variant has not been reported neither in ClinVar nor in LOVD databases. Based on the currently available information, c.1474-1G>T is classified as a likely pathogenic variant according to ACMG guidelines.
