Uncertain significance for Mowat-Wilson syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014795.4(ZEB2):c.388A>G (p.Ile130Val), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 377259). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The valine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with ZEB2-related conditions. This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 130 of the ZEB2 protein (p.Ile130Val). This variant is present in population databases (no rsID available, gnomAD 0.002%).
