Pathogenic — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_007294.4(BRCA1):c.85G>T (p.Glu29Ter), citing Quest Diagnostics criteria: This nonsense variant causes the premature termination of BRCA1 protein synthesis. In addition, it has been reported in individuals and families affected with breast/ovarian cancer in the published literature (PMID: 21233401 (2011), 29446198 (2018)). One study showed this variant caused loss of function in a large-scale study using a haploid cell line (PMID: 30209399 (2018)). This variant has not been reported in large, multi-ethnic general populations. Therefore, the variant is classified as pathogenic.
