Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005670.4(EPM2A):c.736C>T (p.Pro246Ser), citing Ambry Variant Classification Scheme 2023: The p.P246S variant (also known as c.736C>T), located in coding exon 4 of the EPM2A gene, results from a C to T substitution at nucleotide position 736. The proline at codon 246 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
