Submitter rationale: KIF4A, a N5-kinesins highly expressed in differentiated young neurons, moves into the neurites after dissociation from the enzymatic protein poly (ADP-ribose) polymerase 1 (PARP1), which prevents apoptosis (Midorikawa, et al., 2006). Here, it may transports cargoes important for development of neurons, such as the adhesion molecule L1 involved in axon and synapse development (Peretti et al., 2000). Variants involving the KIF4A gene (MIM:300521) are associated in the literature with a clinical condition called “Intellectual developmental disorder, X-linked 100” (MIM:300923) with recessive X-linked transmission mode and loss-of-function mechanism (Kalantari et al., 2021; Willemsen et al., 2014). This condition is characterized by highly variable clinical signs including: developmental delay, convulsions, hydrocephalus, hydranencephaly, ventriculomegaly, cerebellar hypoplasia, agenesis of the corpus callosum, hypoplasia and malrotation of the hippocampus, polymicrogyria, perinodular hyperplasia and interhemispheric cysts. Other anomalies described are those of the kidney and urinary tract, such as dysplastic cystic kidneys. The highlighted variant could interfere with the processing of the gene transcript.
