Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 247320 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.782-3T>A has been reported in the literature in the compound heterozygous state in at least one individual affected with Deficiency Of Aromatic-L-Amino-Acid Decarboxylase (Pearson_2021). These report(s) do not provide unequivocal conclusions about association of the variant with Deficiency Of Aromatic-L-Amino-Acid Decarboxylase. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
