Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000006.11:g.(102372595_102376289)_(102503456_102516221)dup, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant involves the duplication of exons 14-16 in the GRIK2 gene. A presumed nomenclature of c.(1867+1_1868-1)_(2562+1_2563-1)dup has been designated for the purposes of this classification. It is assumed to be a tandem duplication in direct orientation (PMIDs: 25640679, 30054569). This Copy Number Variant (CNV) is expected to alter the reading frame and predicted to result in a truncation. The variant was absent in 21694 control chromosomes in the gnomAD database (Structural Variants v2.1 dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of exon 14-16 duplication in individuals affected with intellectual developmental disorder, autosomal recessive 6 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
