This variant lies in the ALOX12B gene (transcript NM_001139.3) at coding-DNA position 245, where G is replaced by C; at the protein level this means replaces tryptophan at residue 82 with serine — a missense variant. Submitter rationale: Variant summary: ALOX12B c.245G>C (p.Trp82Ser) results in a non-conservative amino acid change located in the PLAT domain (IPR001024) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251352 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.245G>C has been reported in the literature (Sun_2022). This report however, does not provide unequivocal conclusions about association of the variant with Lamellar Ichthyosis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
