This variant lies in the MMUT gene (transcript NM_000255.4) at coding-DNA position 2009, where G is replaced by T; at the protein level this means replaces glycine at residue 670 with valine — a missense variant. Submitter rationale: Variant summary: MUT c.2009G>T (p.Gly670Val) results in a non-conservative amino acid change located in the Methylmalonyl-CoA mutase, C-terminal domain (IPR006159) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250994 control chromosomes. c.2009G>T has been reported in the literature in at least three compound heterozygous individuals affected with Methylmalonic Acidemia (e.g. Liang_2021, Yu_2021, Guo_2024, Ling_2024).
