Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000033.4(ABCD1):c.319C>A (p.Leu107Met), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCD1 c.319C>A (p.Leu107Met) results in a conservative amino acid change located in the ABC transporter integral membrane type-1 fused domain (IPR011527) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 156309 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.319C>A has been reported in the literature in at least an individual affected with adrenoleukodystrophy (example: Nair_2024). These data do not allow any conclusion about variant significance. A different variant affecting the same codon has been classified as likely pathogenic by our lab (c.320T>C, p.Leu107Pro), supporting the critical relevance of codon 107 to ABCD1 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
