Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_018699.4(PRDM5):c.1010G>C (p.Arg337Pro), citing LabCorp Variant Classification Summary - May 2015: Variant summary: PRDM5 c.1010G>C (p.Arg337Pro) results in a non-conservative amino acid change located in the Zinc finger C2H2 type domain (IPR013087) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 250702 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1010G>C in individuals affected with Brittle cornea syndrome 2 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
