Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_003742.4(ABCB11):c.1421C>T (p.Pro474Leu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCB11 c.1421C>T (p.Pro474Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 248762 control chromosomes. c.1421C>T has been reported in the literature at a homozygous state in one individual affected with chronic Intrahepatic Cholestasis (Lipinski_2020). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
