Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004370.6(COL12A1):c.8582G>C (p.Ser2861Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL12A1 c.8582G>C (p.Ser2861Thr) results in a conservative amino acid change located in the Collagen triple helix repeat (20 copies) domain (IPR008160) of the encoded protein sequence. Four of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 248948 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.8582G>C in individuals affected with Ullrich congenital muscular dystrophy 2 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
