Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_173602.3(DIP2B):c.2789A>G (p.Asn930Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: DIP2B c.2789A>G (p.Asn930Ser) results in a conservative amino acid change located in the Dip2 domain (IPR037337) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251442 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2789A>G in individuals affected with Intellectual Disability, FRA12A Type and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
