Submitter rationale: Variant summary: PKD1 c.3814delG (p.Val1272X) results in a premature termination codon, predicted to cause absence of the protein due to nonsense mediated decay, which is a commonly known mechanism for disease. The variant was absent in 238728 control chromosomes. c.3814delG has been reported in the literature in four individuals affected with Autosomal dominant Polycystic Kidney Disease (example, Rossetti_2007). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
