The exact breakpoint at the distal 3' end of this variant is unknown, therefore this deletion may extend downstream of the annotated region of the gene. As it encompasses the termination codon, it is predicted to escape nonsense mediated decay (NMD). The variant was absent in 21694 control chromosomes. c.(2586+1_2587-1)_(*295_?)del has been reported in the presumed heterozygous state in the literature in individual(s) affected with hereditary cancer (LaDuca_2014). These report(s) do not provide unequivocal conclusions about association of the variant with PALB2-related conditions. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. However, at least 1 pathogenic variant in the deleted region (p.Tyr1183*) has been classified as pathogenic by Labcorp, suggesting that loss of these exons is deleterious.
