This variant lies in the DHCR7 gene (transcript NM_001360.3) at coding-DNA position 438 through coding-DNA position 439, replacing the reference sequence with CC; at the protein level this means replaces glycine at residue 147 with arginine — a missense variant. Submitter rationale: Variant summary: DHCR7 c.438_439delinsCC (p.Gly147Arg) results in a non-conservative amino acid change in the encoded protein sequence. Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 279850 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.438_439delinsCC in individuals affected with Smith-Lemli-Opitz Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
