Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_022124.6(CDH23):c.7549A>G (p.Ser2517Gly), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CDH23 c.7549A>G (p.Ser2517Gly) results in a non-conservative amino acid change located in the Cadherins domain profile (IPR002126) of the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 249276 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.7549A>G has been reported in the literature in at least one individual affected with Usher Syndrome (Astuto_2002). These report(s) do not provide unequivocal conclusions about association of the variant with Usher Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
