This variant lies in the PEX13 gene (transcript NM_002618.4) at coding-DNA position 980, where C is replaced by G; at the protein level this means replaces proline at residue 327 with arginine — a missense variant. Submitter rationale: Variant summary: PEX13 c.980C>G (p.Pro327Arg) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251464 control chromosomes (gnomAD). c.980C>G has been reported in the literature in a homozygous individual affected with Zellweger Syndrome (Ebberink_2011). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
