Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001271938.2(MEGF8):c.5347C>T (p.Arg1783Cys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MEGF8 c.5146C>T (p.Arg1716Cys) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 6.7e-05 in 1525498 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in MEGF8 causing Carpenter Syndrome - Type 2 (6.7e-05 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.5146C>T in individuals affected with Carpenter Syndrome - Type 2 and no experimental evidence demonstrating its impact on protein function have been reported.
