Submitter rationale: Variant summary: MEN1 c.80T>C (p.Leu27Pro) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.80T>C has been reported in the literature in at least one heterozygous individual affected with Multiple Endocrine Neoplasia Type 1 and in an additional individual with personal or family history of Multiple Endocrine Neoplasia Type 1 (e.g. Wenzel_2009, Romanet_2019).
