This variant lies in the FANCA gene (transcript NM_000135.4) at 138 bases into the intron immediately before coding-DNA position 2223, where A is replaced by G. Submitter rationale: Variant summary: FANCA c.2223-138A>G is located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: Four predict the variant creates a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 31396 control chromosomes. c.2223-138A>G has been reported in the literature in multiple individuals affected with Fanconi Anemia (Bouchlaka_2003, Ali_2021). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
