This variant lies in the DMD gene (transcript NM_004006.3) at the canonical splice donor site of the intron immediately after coding-DNA position 530, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: DMD c.530+1G>T is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of DMD function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 183350 control chromosomes (gnomAD). c.530+1G>T has been reported in the literature in individuals affected with Dystrophinopathies (e.g. Bennett_2001, Lee_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
