Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000094.4(COL7A1):c.6619G>C (p.Gly2207Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL7A1 c.6619G>C (p.Gly2207Arg) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250822 control chromosomes (gnomAD). p.Gly2207Arg has been reported in the literature in individuals affected with Dystrophic Epidermolysis Bullosa, Dominant (Kon_1997, Varki_2007). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 16971478, 9347800). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
