This variant lies in the MFSD8 gene (transcript NM_001371596.2) at coding-DNA position 1451, where C is replaced by T; at the protein level this means replaces alanine at residue 484 with valine — a missense variant. Submitter rationale: Variant summary: MFSD8 c.1451C>T (p.Ala484Val) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251378 control chromosomes. c.1451C>T has been reported in the literature in a homozygous individual affected with non-syndromic symmetric adult-onset macular dystrophy (Gelormini_2024). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
