Uncertain significance for Hyper-IgE recurrent infection syndrome 3, autosomal recessive — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_001282933.2(ZNF341):c.2496G>C (p.Glu832Asp), citing ACMG Guidelines, 2015: The missense variant c.2496G>C (p.Glu832Asp) in the ZNF341 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes. The amino acid Glutamic Acid at position 832 is changed to a Aspartic Acid changing protein sequence and it might alter its composition and physico-chemical properties. The variant is predicted as damaging by SIFT. The amino acid change p.Glu832Asp in ZNF341 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
