Pathogenic for Cystic hygroma; Fetal megacystis; Branchial cyst; Aberrant right subclavian artery; Thickened nuchal skin fold; Au-Kline syndrome — the classification assigned by Medical Laboratory, The Third Affiliated Hospital of Shenzhen University to NM_031263.4(HNRNPK):c.1074dup (p.Met359fs), citing ACMG Guidelines, 2015: The NM_031263.4: c.1074dup variant in hnRNPK has been found in a fetus of a 30-year-old G2P1 woman. One base duplication in Exon 13 of hnRNPK, which would lead to a translational frameshift. The truncated protein lost an important functional domain (KH Domain Ⅲ) and the truncated protein was predicted to activate nonsense-mediated mRNA decay (NMD). Obstetric ultrasound showed cystic hygroma of the neck, bilateral branchial cysts, megacystis, thickened nuchal fold and Aberrant Right Subclavian Artery‌ (ARSA).
