This variant lies in the CYP11B1 gene (transcript NM_000497.4) at coding-DNA position 61, deleting one base; at the protein level this means shifts the reading frame starting at alanine residue 21, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.61del variant in CYP11B1 led to early termination of translation and a shortened peptide chain(PVS1). The Genome Aggregation Database did not provide information on the approximate frequency of the variant in different populations(PM2_Supporting). Additionally, the affected female fetus presented obvious virilization of the external genitalia(PP4). In summary, the c.61del variant meets our criteria to be classified as likely pathogenic.
