NM_003900.5(SQSTM1):c.1021C>G (p.Leu341Val) was classified as Uncertain significance for Paget disease of bone 2, early-onset; Frontotemporal dementia and/or amyotrophic lateral sclerosis 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SQSTM1 gene (transcript NM_003900.5) at coding-DNA position 1021, where C is replaced by G; at the protein level this means replaces leucine at residue 341 with valine — a missense variant. Submitter rationale: This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 341 of the SQSTM1 protein (p.Leu341Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with amyotrophic lateral sclerosis (PMID: 24138988). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SQSTM1 protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change affects SQSTM1 function (PMID: 27158844, 34929165). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
