Uncertain significance for Familial hypocalciuric hypercalcemia; Autosomal dominant hypocalcemia 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000388.4(CASR):c.965T>C (p.Leu322Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 322 of the CASR protein (p.Leu322Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with hypocalcemia (PMID: 32386559). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects CASR function (PMID: 32386559). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
