Uncertain significance for Dyskeratosis congenita, autosomal recessive 5; Pulmonary fibrosis and/or bone marrow failure, Telomere-related, 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001283009.2(RTEL1):c.3257A>G (p.Tyr1086Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 1086 of the RTEL1 protein (p.Tyr1086Cys). This variant is present in population databases (no rsID available, gnomAD 0.003%). This missense change has been observed in individuals with dyskeratosis congenita (PMID: 30523342). This variant is also known as c.3329A>G (p.Tyr1110Cys). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
