This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 71993, where G is replaced by C; at the protein level this means replaces arginine at residue 23998 with proline — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 23998 of the TTN protein (p.Arg23998Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive congenital myopathy (PMID: 30365001). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant is located in the A band of TTN (PMID: 25589632). Variants in this region may be relevant for cardiac or neuromuscular disorders (PMID: 25589632, 23975875).
