Uncertain significance for Primary open angle glaucoma; Glaucoma 1, open angle, E; Amyotrophic lateral sclerosis type 12 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001008212.2(OPTN):c.307G>A (p.Glu103Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 103 of the OPTN protein (p.Glu103Lys). This variant is present in population databases (rs759022584, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with OPTN-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt OPTN protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
