Uncertain significance for Epilepsy, idiopathic generalized, susceptibility to, 13; Epilepsy, childhood absence 4; Idiopathic generalized epilepsy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001127644.2(GABRA1):c.500C>T (p.Pro167Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 167 of the GABRA1 protein (p.Pro167Leu). This variant is present in population databases (rs750728250, gnomAD 0.01%). This missense change has been observed in individual(s) with clinical features of GABRA1-related conditions (PMID: 33057194, 35982159). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GABRA1 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
