Uncertain significance for Pulmonary fibrosis and/or bone marrow failure, Telomere-related, 3; Dyskeratosis congenita, autosomal recessive 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001283009.2(RTEL1):c.2021_2025+27dup, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RTEL1 gene (transcript NM_001283009.2) at coding-DNA position 2021 through 27 bases into the intron immediately after coding-DNA position 2025, duplicating this region. Submitter rationale: This sequence change falls in intron 23 of the RTEL1 gene. It does not directly change the encoded amino acid sequence of the RTEL1 protein. This variant is present in population databases (no rsID available, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with RTEL1-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the effect of this variant on mRNA splicing is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
